Description

The Leukodif 200 kit is intended for rapid staining of blood smears.

Method principle:

Contains solutions for rapid staining of blood smears. For staining, blood smears are immersed in staining solutions. The intensity of the staining is controlled by the number of dye immersions.

Packing and reagents:

• R1: Fixing solution: methanol 24 mol / l, naphthalene green 1 mg / l (1 vial x 200 ml);
• R2: Staining solution 1: Eosin U 1.73 mmol / L, phosphate buffer pH 6.8 60 mmol / L (1 bottle × 200 ml);
• R3: Staining solution 2: Azur II 12 g / L, phosphate buffer pH 6.8 60 mmol / L (1 bottle × 200 ml);
• R4: Tablets for preparation of washing solution: phosphate buffer pH 7.2 2.5 mmol / table (5 tablets).

Preparation of rinsing solution R4:

One tablet is dissolved in 250 ml of distilled water.

Stability: 2 months at temperatures from +2 to +25 ° C in a dark place.

Progress:

• Blood smears prepared on defatted slides are left to air dry.
• The solutions are poured into vessels.
• The smear is fixed by immersion 5 times for 1 second in reagent R1. After each immersion, the solution is allowed to drain and the excess is removed from the rim of the vessel.
• The fixed swab is immersed 3 times for 1 second in reagent R2. After each immersion, the solution is allowed to drain and the excess is removed from the rim of the vessel.
• The swab is immersed 6 times for 1 second in reagent R3. Also, after each immersion, the solution is allowed to drain and its excess is removed from the edge of the vessel.
• The slides are treated with a washing solution and allowed to air dry.

Warning:

• The color intensity can be adjusted by the number of dives. Do not immerse the smear less than 3.
• The intensity of eosinophilic staining is controlled by the number of immersions in the reagent R2.
• The intensity of basophilic staining is controlled by the number of immersions in the reagent R3.
• The solutions are stable, but should be protected from direct sunlight. If the color of the solution differs from the standard, then the solutions should be replaced. The solutions are usually changed once a week.
• If the calculation of leukocytes turns out to be pathological, then staining should be carried out using solutions according to May-Günwald and Romanovsky-Giemsa. When using the Leukodif set, less intense staining of basophil granulation. Staining is not recommended for platelet testing.

Precautionary measures:

The reagent kit is intended for in vitro diagnostics by a professionally trained laboratory technician.

Reagent 1 contains methanol (100%), which is a poisonous and flammable substance.
