Pharmacological properties
Non-steroidal anti-inflammatory drug with analgesic and antipyretic properties.
Pharmacodynamic action
Ketoprofen belongs to the generation of non-steroidal anti-inflammatory drugs based on propionic acid.
The inflammatory agent damages the cell and activates the phospholipidases that supply arachidonic acid. It is the basis for the enzyme systems of cyclooxygenase and lipoxygenase, which result in the formation of substances that provoke inflammation: prostaglandins, thromboxanes and leukotrienes. Ketoprofen is a dual inhibitor of inflammation, blocking the action of cyclooxygenase and lipoxygenase and, thus, preventing the formation of prostaglandins and leukotrienes.
In addition, ketoprofen is an analgesic that affects the central and peripheral nervous system. Its action is to directly suppress the action of bradykinin, a vasodilator and pain mediator.
In addition to acting against bradykinin, ketoprofen is active in the central nervous system, suppressing pain.
In horses, ketoprofen neutralizes the effect of endotoxins and counteracts intestinal spasms caused by bradykinin.
Pharmacokinetic data
Ketoprofen is rapidly absorbed. In cows, peak plasma levels (8.025 ± 1.9 μg / ml) are reached less than an hour after intramuscular administration, and the bioavailability of the substance is almost complete. More than 98% binds to plasma proteins and accumulates in inflamed tissues. Ketoprofen has a high affinity for synovial tissues, spreads rapidly and is excreted. The duration of action of the drug is longer than expected, according to its half-life, which lasts from less than one hour (intravenous administration to horses) to more than five hours (after intramuscular administration to cattle).
Significant concentrations of ketoprofen were found in the synovial fluid and its level is higher than in plasma, and the half-life is 2-3 times higher than in plasma. Ketoprofen is metabolized in the liver to insignificant biologically active metabolites, and 90% is excreted in the urine as glucuronide compounds.
Application
Cattle: as an anti-inflammatory, analgesic and antipyretic agent for the treatment of inflammatory processes caused by respiratory diseases, edema of the mammary glands, acute mastitis, inflammatory processes in the musculoskeletal system.
Horses: treatment of inflammatory and painful conditions in diseases of the musculoskeletal system (lameness of traumatic origin, arthritis, osteoarthritis, joint injuries (sprains, synovitis), fractures, tendinitis, hoof lesions (pododermatitis, lameness), inflammation after surgery) ; symptomatic treatment of colic.
Pigs: anti-inflammatory, analgesic and antipyretic therapy for MMA syndrome, treatment of inflammatory and painful conditions in diseases of the musculoskeletal system; in respiratory infections.
Dosage
Cattle : intravenously or intramuscularly 3 ml of the drug per 100 kg of body weight (3 mg of ketoprofen / kg of body weight) once a day for 1-3 days.
Horses: intravenously. For the treatment of lesions of bones and joints, as well as the musculoskeletal system, inject 1 ml of the drug per 45 kg of body weight (2.2 mg of ketoprofen / kg of body weight) once a day for 3-5 days. For symptomatic treatment of colic, inject 1 ml of the drug per 45 kg of body weight (2.2 mg of ketoprofen / kg of body weight) once.
Additional injections are performed only after re-clinical examination of the animal by a veterinarian.
Pigs: intramuscularly 3 ml of the drug per 100 kg of body weight (3 mg of ketoprofen / kg of body weight) once a day for 1-3 days.
Contraindication
Contraindicated in animals with severe renal insufficiency.
Do not administer to animals sensitive to ketoprofen.
Reservation
After intramuscular injection, local edema may appear, painful to the touch, which may swell up to 10 cm in radius and disappear a week after treatment.
Foals for mares and foals under 15 days of age should not be administered.
Do not administer other NSAIDs, diuretics or anticoagulants at the same time.
Consumption of milk for human consumption is allowed without restrictions
Slaughter of animals for meat is allowed 4 days after the last administration of the drug.
