Composition and form of release
The drug in the form of a white or light yellow lyophilized powder for injection complete with a solvent (water for injection) is produced in glass vials sealed with chlorobutanol stoppers and rolled up with aluminum caps.
Each bottle contains as an active ingredient: 250mg of tiletamine hydrochloride and 250mg of zolazepam hydrochloride (Zoletil 100). Excipients – sodium sulfate and lactose.
Pharmacological properties
Tiletamine is a general anesthetic of dissociative action, causing a pronounced analgesic effect, but insufficient muscle relaxation. Tiletamine does not suppress the pharyngeal, laryngeal, cough reflexes, does not depress the respiratory system.
Zolazepam inhibits the subcortical areas of the brain, causing anxiolytic and sedative effects, relaxes the striated muscles. Zolazepam enhances the anesthetic effect of tiletamine. It also prevents tiletamine-induced cramps, improves muscle relaxation, and speeds up recovery from anesthesia.
The time of the onset of anesthesia after intramuscular administration is individual, therefore, after the injection of the drug, careful observation of the animal should be established.
In cats, the onset of action of the drug after intramuscular administration is noted after 1-7 minutes. The duration of anesthesia depends on the dose, but the maximum effect is usually observed after 20-60 minutes. The recovery period after anesthesia varies from 1 to 5.5 hours.
In dogs, the onset of action of the drug after intramuscular administration is noted on average within 7.5 minutes. The average duration of anesthesia is about 30 minutes, the recovery period is up to 4 hours.
In case of overdose, as well as in very young and old animals, the recovery period is longer. Zoletil’s active ingredients are metabolized in the liver and excreted in the urine.
Indications for use
General anesthesia for diagnostic and surgical procedures in cats and dogs.
The order of application and dose
24 hours before anesthesia, the antiparasitic collar should be removed from the animal and a 12-hour fasting diet should be maintained.
15 minutes before the administration of the drug, it is recommended to inject atropine sulfate subcutaneously at a dose of: dogs – 0.1 mg / kg of animal weight, cats – 0.05 mg / kg of animal weight.
Dilute the contents of the powder bottle with the supplied solvent. With intramuscular administration, the loss of rectifying reflexes occurs after 3-6 minutes, with intravenous administration – after 1 minute.
Intramuscular injection:
Dogs: clinical examination – 7-10mg / kg; short-term general anesthesia for minor surgical interventions – 10-15 mg / kg; long-term general anesthesia for extensive and painful surgical interventions – 15-25 mg / kg.
Cats: clinical examination – 10mg / kg; long-term general anesthesia for extensive and painful surgical interventions – 15 mg / kg.
Intravenous administration:
Dogs: clinical examination – 5mg / kg; short-term general anesthesia for minor surgical interventions – 7.5 mg / kg; long-term general anesthesia for extensive and painful surgical interventions – 10 mg / kg.
Cats: clinical examination – 5 mg / kg; long-term general anesthesia for extensive and painful surgical interventions – 7.5 mg / kg.
If necessary, zoletil can be re-administered at a dose not exceeding 1 / 3-1 / 2 of the initial dose.
Zoletil does not have a cumulative effect and can be re-injected in doses not exceeding 1 / 3-1 / 2 of the initial dose. In this case, the total dose of the drug should not exceed the safety threshold: 30 mg / kg for dogs and 72 mg / kg for cats; in both animal species, the minimum lethal dose is 100 mg / kg. The duration of anesthesia is from 20 to 60 minutes.
Recovery from anesthesia: The analgesic effect of Zoletil is longer than that caused by surgical anesthesia. The exit from anesthesia is gradual (2-6 hours) and calm, provided there is no noise and bright light. In cases of overdose, as well as in very young and old animals, the recovery period is longer.
Contraindications
Zoletil is contraindicated in animals with diseases of the pancreas, with severe disorders of the cardiovascular and respiratory systems, hypertension. For animals with kidney disease, the dose should be reduced. Zoletil crosses the placenta and may cause respiratory depression in the fetus.
Side effects
With the introduction of high doses or intolerance to zoletil, apnea may occur. During awakening, vomiting, excessive salivation and bronchial secretion (if premedication with atropine has not been carried out), involuntary muscle twitching, hypertension, muscle stiffness can be observed.
In case of an overdose, artificial lung ventilation is performed. Other symptoms are treated with symptomatic therapy.
special instructions
The simultaneous use of zoletil and chloramphenicol prolongs anesthesia in cats by an average of 30 minutes.
The simultaneous use of zoletil with phenothiazine (acepromazine) causes respiratory and cardiac depression. With the simultaneous use of zoletil with barbiturates or inhalation anesthetics, the dose of the latter should be reduced.
