Pharmacological properties

Ceftiofur belongs to the third generation of cephalosporin antibiotics. The drug has a broad spectrum of antibacterial activity and is active against gram-positive and gram-negative microorganisms, including strains that produce β-lactamase and some types of anaerobic bacteria: Pasteurella haemolytica, Pasteurella multocida, Actinobacillus (Haemophilus, Haemophilus melaninogenicus, Actinobacillus pleuropneumoniae, Pasteurella multocida, Streptococcus suis, Streptococcus zooepidemicus, Streptococcus equi, Staphylococcus ssp., Pasteurella ssp., Escherichia coli, Staphylococcus aureus. The antibacterial action of ceftiofur is realized by the mechanism of inhibition of bacterial cell wall synthesis. Following parenteral intramuscular administration, ceftiofur is rapidly absorbed and rapidly metabolized to form furoic acid and desfuroylceftiofur. Maximum concentrations of ceftiofur and its metabolites in blood plasma are observed after 30-120 minutes and are maintained at therapeutic levels for up to 24 hours after administration of the drug. The activity of the drug is not reduced in the presence of necrotic tissue. High levels of protein-bound ceftiofur in adult animals affect its distribution, which distinguishes it from other cephalosporins. The primary metabolite of ceftiofur, desfuroylceftiofur, is a reactive sulfhydryl group that forms bilateral covalent bonds with tissue proteins and blood plasma. Free concentrations of ceftiofur and its metabolites are usually lower than the doses administered, which is probably due to the property of active metabolites to bind to proteins. The kidneys, under conditions of their normal functional state, within 24 hours excrete approximately 95% of the intramuscular dose of the drug, about 60-80% is excreted in the urine by glomerular filtration and through the secretion of renal tubules. Most of the excreted dose is excreted in the form of desfuroylceftiofur and desfuroylceftiofur cysteine disulfide with a small amount of non-metabolized ceftiofur.
Application
Cattle: treatment of animals with respiratory diseases caused by Pasteurella (Mannheimia) haemolytica, Pasteurella multocida, Histophilus somni, as well as animals with acute interdigital necrobacilliosis caused by Fusobacterium necrophorum, which are sensitive to Bactero.
Pigs: treatment of animals with respiratory diseases caused by Actinobacillus (Haemophilus) pleuropneumoniae, Pasteurella multocida, Salmonella choleraesuis and Streptococcus suis, which are sensitive to ceftiofur.
Horses: treatment of animals with respiratory diseases caused by Streptococcus zooepidemicus that are sensitive to ceftiofur.
Dogs: treatment of animals with polyarthritis, as well as respiratory, urinary tract and skin diseases caused by Escherichia coli and Proteus mirabilis, which are sensitive to ceftiofur.
Day-old chicks and turkeys: prevention of early mortality caused by E. coli and Staphylococcus aureus, which are sensitive to ceftiofur.
Dosage
The contents of one vial are dissolved in 20 ml of sterile water for injections. 1 ml of the prepared standard solution contains 50 mg of ceftiofur in the form of sodium salt.
The shelf life of the resulting solution is: 12 hours when stored in a dark place at a temperature of 15 to 30 ºC, when stored in a refrigerator at a temperature of 2 to 8 ºC – 7 days.
When stored in a frozen state at a temperature of minus 4 to minus 18ºC – 8 weeks.
Defrost the frozen solution at 37 ºC and do not freeze it a second time.
Intramuscularly in a dose:
cattle – 1 ml of drug solution per 50 kg of body weight (1 mg of ceftiofur per 1 kg of body weight). No more than 15 ml of sterile solution of the drug can be injected into the injection site.
The drug is used once a day for 3-5 days – in the treatment of respiratory diseases, and in the treatment of interdigital necrobacillosis – for 3 days;
pigs – 1 ml of drug solution per 16 kg of body weight (3 mg of ceftiofur per 1 kg of body weight) once a day for 3 days;
horses – 2 mg of ceftiofur per 1 kg of body weight (1 ml of drug solution per 25 kg of body weight) 1 time per day for 10 days with an interval of 24 hours between injections. No more than 10 ml of sterile solution of the drug can be injected into the injection site. Treatment is continued for 48 hours after the disappearance of clinical signs of the disease, but not more than 10 days.
Subcutaneously in a dose:
dogs – in the treatment of respiratory diseases – 0.6 ml of drug solution per 10 kg of body weight (3.3 mg of ceftiofur per kg of body weight) for 5 days; in diseases of the urinary tract – 0.8 ml of the drug solution per 10 kg of body weight (4.4 mg of ceftiofur per kg of body weight) for 8-10 days. In skin diseases, the dose is 0.4-0.8 ml of the drug solution per 10 kg of body weight (2.2-4.4 mg of ceftiofur per kg of body weight) for 10 days. At purulent wounds the dose makes 0,6 ml of a solution of drug on 10 kg of body weight (3,3 mg of ceftiofur on 1 kg of body weight) within 8-10 days. In the treatment of polyarthritis, the drug is administered in a dose of 0.8 ml of drug solution per 10 kg of body weight (4.4 mg of ceftiofur per 1 kg of body weight) for 8 days. Dosage and duration of treatment depends on the severity of the disease;
day-old chicks – 0.2 ml of the drug solution per head (in the neck); Before use, the standard solution of the drug (50 mg / ml) is diluted in such a way that 0.2 ml of the solution contained 0.08-0.2 mg of ceftiofur per chicken;
one-day-old turkeys – in a dose of 0.2 ml of the drug solution per head (in the neck); Before use, the standard solution of the drug (50 mg / ml) is diluted in such a way that 0.2 ml of the solution contained 0.2-0.5 mg of ceftiofur per chicken (1 ml of drug solution per 100 – 294 heads).
Contraindication
Do not use in animals with hypersensitivity to ceftiofur or other beta-lactam antibiotics.
Reservation
The drug is not prescribed to animals if they are aware of their individual intolerance to cephalosporins. Topical exposure to ceftiofur may cause allergic reactions in susceptible individuals. People with known hypersensitivity to penicillins and cephalosporins should avoid contact with the drug. Repeated or prolonged exposure to ceftiofur may cause sensitization. Avoid contact with clothing, skin, eyes and mouth. In case of accidental contact with eyes, rinse immediately with water for 15 minutes. In case of accidental contact with the skin, wash with soap, get rid of contaminated clothing. If after contact with the drug there is an allergic reaction (eg, skin rash, urticaria, shortness of breath) should consult a doctor immediately.
Slaughter of animals for meat is allowed after 24 hours (cattle), 48 hours (pigs), 21 days (chickens, turkeys) after the last application of the drug. The excretion period for milk is 0 hours. The meat obtained before this deadline is disposed of or fed to unproductive animals, depending on the opinion of the veterinarian.
