Covinan in 1 ml contains as an active ingredient 100 mg of proligestone, which is a synthetic analogue of progesterone, as auxiliary substances: methyl-, propyl parahydroxybenzoate, sorbitan palmitate, polysorbate 40, lecithin, sodium citrate, potassium dihydrogen phosphate, macrogol 4000, water for injection.
Pharmacological properties:
Proligestone is a steroid hormone that inhibits the secretion of gonadotropic hormones (especially luteinizing hormones), inhibits the maturation of follicles in the ovaries, blocks the release of estradiol from mature follicles and causes their regression, which prevents the development of estrus. Proligestone causes changes in the mucous membrane of the uterus and increases the viscosity of mucus in the uterus, which prevents the implantation of a fertilized egg, inhibits cell hyperplasia and the secretion of mammary glands in animals. When administered during the anestrus phase, proligestone maintains a low LH concentration in the body, which prevents follicular growth and provides contraception for the animal. When the drug is discontinued, the concentration of proligestone in the body decreases, restoring the normal level of LH in the animal’s blood, and the contraceptive effect of the drug stops. Proligestone has very weak estrogenic, negligible corticosteroid activity and has no androgenic effect. The prolonged action of the drug is associated with the deposition of proligestone in adipose tissue and circulation in the liver. Dose and method of administration:
Covinan is injected strictly subcutaneously, in the middle part of the skin fold, avoiding intradermal injections, as well as injections into subcutaneous fat deposits, scar tissue. Before administration, the drug is thoroughly shaken, the injection site is thoroughly disinfected with 70% alcohol solution. For the prevention and treatment of false pregnancy and pseudolactation in bitches, the drug is administered once in a dose of 1 -, 3 ml, depending on the breed and weight of the animal: up to 20 kg -, 1 ml, 21 -, 40 kg -, 2 ml, over 40 kg – , 3 ml). To suppress sexual desire in cats, Covinan is used in a dose of 1 ml per animal (for large cats weighing more than 7 kg, the dosage can be increased to 1.5 ml). For bitches, the dose is selected individually (from 10 to 33 mg / kg), depending on the weight of the animal:

For the development of persistent anestrus in bitches and cats, Covinan is used in the indicated doses according to the basic scheme:
first injection during anestrus (for bitches and cats) or proestrus (for bitches),
second injection 3 months after the first injection,
the third injection 4 months after the second injection.
then the drug is administered every 5 months.
If the bitches, who are using Covinan according to the indicated scheme, show signs of proestrus (precursor), in order to prevent unwanted estrus, it is necessary to make an extra injection of the drug in the appropriate dose.
The further scheme of use depends on the interval with which the injections were made before the appearance of undesirable signs of proestrus:
if the interval between the next injections was 5 months, then, after an extraordinary injection, Covinan is administered after 4 months, and then – every 5 months:
if the interval was 4 months, then after an extraordinary injection, further Covinan is administered according to the main scheme (after 3 months, then after 4 months, then the drug is injected every 5 months).
For a single prevention of sexual heat in females and cats, Covinan is administered once, for females – during the anestrus period, preferably no later than a month before the expected estrus, for cats -, Covinan is administered during the di- or anestrus period.
To suppress estrus in bitches, Covinan is administered immediately after signs of proestrus appear (spotting and loop enlargement). Signs of proestrus gradually disappear within 5-7 days.
The restoration of the reproductive cycle in bitches occurs on average after 6-7 months; in rare cases, estrus is restored in the interval from 3 to 12 months.
When using the drug in rare cases (0.1 – 0.6% of cases), side effects such as pyometritis, endometrial hyperplasia, a slight increase in body weight, increased appetite, apathy, and slight hypertrophy of the mammary glands are possible. The likelihood of side effects increases if the animals are simultaneously used other progesterone analogues. A minor local painful reaction is possible immediately after administration of the drug. At the injection site, lightening of the coat and slight hair loss are possible. Like other progestogens, the drug can cause adrenal suppression.
Contraindications:
Covinan is not used in pregnant and lactating animals, during estrus, animals with diseases of the genitourinary system, animals with prolonged vaginal discharge, bitches who received estrogens and progestogens for the treatment of false gestation, and it is also undesirable to administer to bitches before the first estrus and in the later stages of proestrus. For diabetic animals, the drug is used with careful monitoring of blood glucose levels.
